DOCK2 regulates platelet biogenesis and functions by controlling cytoskeletal actin dynamics

The regulation of cytoskeletal actin dynamics is critical for platelet biogenesis and activation. In this presentation, Chao Fang explores the role of DOCK2, a Rho GTPase regulator, in platelet biology. Using DOCK2-deficient mice, the study observed macrothrombocytopenia and altered actin dynamics. DOCK2 deficiency led to reduced Rac1 activity, impaired GPVI-mediated platelet activation and defective thrombus formation. Pharmacological inhibition of DOCK2 similarly disrupted platelet function. These findings suggest that DOCK2 is essential for platelet formation and stability and may represent a potential therapeutic target in thrombotic and thrombo-inflammatory diseases.
